What Is Prolia?

Prolia® (denosumab) is an FDA-approved biologic injection used to strengthen bones and reduce fracture risk in patients with:

  • Postmenopausal osteoporosis
  • Osteoporosis in men
  • Bone loss due to hormone-depleting cancer treatments

Prolia works by inhibiting RANKL, a protein involved in bone breakdown, thereby reducing bone resorption and increasing bone density.

It is administered as a subcutaneous injection every 6 months by a trained healthcare provider.

Who Should Receive Prolia?

Prolia may be prescribed for:

  • Women with postmenopausal osteoporosis at high risk for fracture
  • Men with osteoporosis
  • Adults experiencing bone loss due to prostate or breast cancer treatment
  • Patients unable to tolerate or unresponsive to oral bisphosphonates

Your provider will determine if Prolia is appropriate based on bone density scans (DEXA), fracture history, and treatment goals.

Prolia Side Effects

Prolia is generally well tolerated, but potential side effects include:

  • Back, arm, or leg pain
  • Muscle or joint pain
  • High cholesterol
  • Low calcium levels (especially in patients with kidney disease)
  • Skin infections (rare)
  • Jaw problems (osteonecrosis, rare with long-term use)

Calcium and vitamin D supplements are often recommended alongside Prolia to support bone health.
